ARP not resolve

HI,
i have tested my ethernet application with my IIS server and ARP request is acknowledged but when i connect it to main server and try to access a webpage but as i changed the ip address to desired one and port to desired value (80) suddenly ARP is not having acknowledgment from server.
if some one might had this problem or like to help me, i shall be thankful.
